Oops, I see I missed a point that has you on I-64 for a short bit, and I started you at Hubbard, that you can alter easy enough.
Enjoy the ride, I am jealous, I too would like to stay down there!

Take the Blue Ridge Parkway, it will spit you out in Cherokee, just around the mountains from 129. Cut over Foothills Parkway and you are there.

The 5 or 7 times we've taken that route we ride US33 in Ohio through WV and into VA (very nice once the 4 lane ends in Ohio), 33 takes you right to the north end of the BRP, and then ride it on down for 400+ miles of twisting two lane with no stop sign or red light to be seen.

You can easily ride this route in 2 days.

I'll second the BRP, but since you're coming from the NE I'd head down to Wheeling, WV to catch US-250 and take it to Elkins, WV where you'll run into US-33 and SR-55. The two split at Seneca rocks; SR-55 will take you to Front Royal, VA and the north end of Skyline Drive, and as Meef noted US-33 will dump you out at the north end of the BRP near Elkton, VA.
